Inhalation toxicology has traditionally leaned on epithelial cell models for alveolar safety assessment. While these models provide insights into barrier function and cytotoxicity, they may not comprehensively address the multifaceted inflammatory responses integral to long-term lung health, especially in the context of chronic exposure.
The sentinel role of alveolar macrophages, the lung’s immediate responders, cannot be overstated. They are not just indicative of immediate reactions but also offer a window into the broader health ramifications within the respiratory system. At ImmuONE, we recognise the significance of this complex interplay. Hence, we champion the use of immune-competent models, focusing on the intricate dynamics of alveolar macrophages. By doing so, we endeavour to offer a more holistic understanding of the inflammatory impact and overall safety of inhaled substances.

Atherosclerosis
Macrophages play a central role in the development of plaques found in atherosclerotic cardiovascular disease. Classically activated pro-inflammatory macrophages are implicated in initiating and sustaining plaques while alternatively activated anti-inflammatory macrophages are linked to the resolution of inflammation and plaque stabilisation.
We are currently developing lipid_ONE, an assay designed to predict how the intracellular lipid profiles of macrophages alter with compound exposure. lipid_ONE could be used to predict whether exposure to a substance contributes to the initial events that lead to atherosclerosis.
